On-chip staining without cell loss

The diameter of pores in filter is 2 μm. Only fluid is allowed to pass through the pores. Captured cells are kept in microwells during staining and washing.

You can stain race cells like circulating tumor cells, fetal cells etc. without cell loss during fixation, permeabilization, blocking, incubation and washing.


Example

A549 human lung adenocarcinoma cell is EpCAM low expression cell line.

Step 1

Loading of A549 cells.

Step 2

FITC labelled mouse anti-human EpCAM mAb was added.

Step 3

Excess reagent was washed after 30 minutes incubation. The fluorescent intensity is very weak.

Step 4

Alexa® 488 labelled goat anti-mouse IgG was added.

Step 5

Excess reagent was washed after 30 minutes incubation. 2nd staining increases the fluorescence intensity.
